Jean-Paul Duminy took the highest 51 off 99 balls while David Andrew Miller 44 off 57 balls.
World’s top all-rounder Shakib-Al-Hasan bagged three wickets.
He touched the landmark of the 200-wicket club in the match as second Bangladeshi after Abdur Razzak.
Besides, Rookie Mustafizur Rahman and Rubel Hossain bagged two wickets each while Mahmudullah and Mashrafe Morataza picked up one each.
Earlier, South Africa won the toss and elected to bat first. The match began at 3:00 local time (09:00 GMT).
Morne Morkel replaces Chris Morris while Bangladesh are unchanged.
Hometown boy Tamim Iqbal has the most runs (391) at this ground, while Shakib Al Hasan has the most wickets (27).
